PURPOSE: An anticancer agent containing Hades as a Akt negative regulator is provided to suppress proliferation and presence of cancer cells. CONSTITUTION: An Akt(protein kinase B) negative regulator contains Hades protein with an amino acid of sequence number 2 or a gene encoding the same as an active ingredient. The gene encoding Hades contains a base of sequence number 1. The gene is inserted into a recombinant expression vector. Hades contains RING finger-domain and has E3-ubiquitin ligase activity. An anticancer agent contains Hades protein or the gene encoding the same as an active ingredient. The anticancer agent functions as an Akt(protein kinase B) negative regulator.본 발명은 Akt 음성 조절제로서의 Hades의 용도에 관한 것으로, 더욱 구체적으로 Hades 단백질 또는 이를 코딩하는 유전자를 유효성분으로 함유하는 Akt (단백질 키나아제 B) 음성(negative) 조절제 또는 항암제에 관한 것이다. 세린/트레오닌 키나아제인 Akt 는 세포 증식(proliferation), 생존(survival) 및 종양(tumor) 발달을 포함한 다양한 세포 프로세스들에서 기능한다. 본 발명자들은 RING 핑거 도메인 및 E3 유비퀴틴 리가아제 활성을 갖는, Akt 의 음성 조절인자인 Hades를 동정하였다.본 발명에 따르면, Akt 는 Hades와 직접적으로 상호작용하여, Hades에 의해 시험관내(in vitro) 및 생체내(in vivo)에서 유비퀴틴화된다. 따라서, 본 발명에 따른 Hades는 Akt를 분해하고 그 다운스티림 시그날링을 음성 조절함으로써 암세포의 증식 및 생존을 억제할 수 있으므로, 항암제로 유용하게 사용될 수 있다.
