PROBLEM TO BE SOLVED: To provide a method for forming a Matsutake (Tricholoma matsutake) fungus-infected seedling by cocultivating a Matsutake fungus strain and an aseptically germinated pine seedling. SOLUTION: In this method for forming the Matsutake fungus-infected pine seedling by cocultivating a Matsutake fungus and the aseptically germinated pine seedling, a mycelium powdered material which is obtained by powdering a fruit body of the Matsutake fungus given by subjecting the fungus to liquid culture in a PDB (potato dextrose broth) culture medium is inoculated in an amount in a dry base of 0.01-0.02mg per 1ml of sterile water on the bottom of a sterilized culture vessel. Then, a K-liquid culture medium which is specifically formulated and controlled to have a pH of 5.5 to 5.6 is dividedly injected into mixed bed soil which is obtained by mixing perlite and sphagnum peat moss in a ratio of 80 to 1-2 and arranged on the inoculated mycelium powdered material. Further, the culture vessel is covered with a lid, after the aseptically germinated seedling which is obtained by aseptically geminating a pine seed and has a length of≤3cm is laid on an infection culture medium containing the mixed bed soil and the K-liquid culture medium and embedded therein. Furthermore, the pine nersery seedling and a Matsutake fungus culture are cocultivated at 15-25°C for 24 hr in a lighting condition of 10-40,000lux. COPYRIGHT: (C)2004,JPO
