Disclosed is a chimeric flavivirus comprising a yellow fever virus of strain YF17D in which the membrane and envelope proteins of the yellow fever virus are replaced with the membrane and envelope proteins of West Nile virus strain NY99, wherein said chimeric flavivirus comprises the following mutations: (a) the substitutions L107F, A316V and K440R in the envelope protein； (b) the C2 deletion (deletion of amino acids PSR, residues 40-42) in the capsid protein； and additionally either: (c)(i) a deletion in the 3&rsquo； untranslated region (3&rsquo； UTR) of the chimeric flavivirus genome selected from: dB (deletion of CAGGT, nucleotides 256-260), dD (deletion of CGGAG, nucleotides 308-312) and d7 (deletion of AAGACGG, nucleotides 345-351)； or (c)(ii) the substitutions Y176V, T177A and K280M in the envelope protein.
