Gout is a common one of metabolic diseases in Taiwan. With the westernization of the national diet,cause Hyperuricemia and Gout gradually increased and getting younger and younger. Our invention use MSU to induce RAW 264.7 cell that xanthine oxidase has high active in cell. After RAW 264.7 cell treatment with miracle fruit water extract which xanthine oxidase active decrease along of concentration increases. When the concentration of 500 μg/mL, xanthine oxidase active reply to control. And it effect better than Allopurinol (100 μg/mL). Miracle fruit water extract has more effect when it together with vitamine C (100 μg/mL). This is result confirm miracle fruit water extract can inhibit xanthine oxidase active, and then reduce uric acid in blood. Our invention also confirm it can scavenging reactive oxygen species. In summary, miracle fruit water extract not only inhibit xanthine oxidase active, but also scavenging reactive oxygen species. Miracle fruit is safety to eat and no side effect that can as patient’s health food in diet.目前痛風在台灣屬一種常見之代謝性疾病,許多流行病學研究皆顯示現代化的生活形態導致亞洲人的高尿酸血症及痛風關節炎的盛行率持續增加,且有年輕化之趨勢。本發明以尿酸鹽結晶誘發RAW 264.7細胞造成胞內黃嘌呤氧化酶具高活性,以神秘果水萃物處理細胞後,酵素活性隨樣品濃度增加而降低,當神秘果水萃物濃度為500 μg/mL,含5.39 μg酚類化合物時,黃嘌呤氧化酶活性已回復與控制組相似,效果較正對照組異嘌呤醇(100 μg/mL)佳,且與維生素C(100 μg/mL)共存時具有加乘效應,證實本發明之組合物可有效抑制黃嘌呤氧化酶之活性,進而降低血液中尿酸含量。經實驗證實,神秘果水萃物亦具有清除活性氧分子之功效,此結果說明神秘果水萃物不僅可抑制黃嘌呤氧化酶活性,亦可清除活性氧分子,具有預防及治療高尿酸血症之潛力,且神秘果食用安全無副作用,可作為患者日常食用之保健產品。
