Molecular determinants of cancer response to immunotherapy are described, and systems and tools for identifying and / or characterizing cancers that are likely to respond to immunotherapy are also described. The present invention encompasses the discovery that the potential for a favorable response to cancer immunotherapy can be predicted. The present invention further includes the discovery that cancer cells may carry somatic mutations that result in neoepitope that can be recognized by the patient's immune system as non-self.
