Systems and methods are provided for separating fluid containing red blood cells into separated red blood cells and other fluid components. Suitable systems include a disposable fluid flow circuit and a durable, reusable separation system that is mounted on or associated with the separation system. The circuit includes a membrane separator for separating fluid into components and a leukocyte reduction filter. The leukocyte reduction filter can be used before or after fluid containing red blood cells is passed through the membrane separator.
