DNA ENCODING CYSTEINE PROTEINASE DERIVED FROM WHEAT SEED, TRANSFORMED E. COLI CONTAINING THE DNA AND RECOMGINANT CYSTEINE PROTEINASE EXPRESSED BY THE E. COLI
PROBLEM TO BE SOLVED: To obtain the subject DNA useful for obtaining by genetic engineering techniques wheat seed cystein proteinase which is expected to have a wide variety of uses as food enzymes and pharmaceutical enzymes. SOLUTION: This DNA is derived from wheat seed, and is composed of the base sequence from No.62 to No.1192 in the base sequence of the formula or encodes the proteins which is composed of the amino acid sequence of the formula and has a cystein proteinase activity. The DNA is obtained by making a cDNA library from the mRNA, as a template, extracted from wheat seed during germination, and by screening according to the Southern blot technique and subcloning where cDNA fragments containing an encoded region of oryzaninα,βorγof rice seed cystein proteinase are used as probes, followed by determining the base sequence and the amino acid sequence. The DNA is also obtained by producing the plasmid for expression of recombinant proteins, by producing a transformant and then by culturing the transformant.
