The present invention is based on confirmation that a hepatitis B virus antibody interferes with a surface antigen of hepatitis B virus bonded to heparan sulfate proteoglycan, thereby suppressing the formation of cccDNA of hepatitis B virus. A pharmaceutical composition and a method for suppressing the formation of cccDNA of hepatitis B virus of the present invention can be used to fundamentally treat chronic hepatitis B, and can be expected to prevent reoccurrence of hepatitis following liver transplantation in a hepatitis B patient. Additionally, according to the present invention, novel substances for preventing or treating hepatitis B can be identified by confirming suppression of the cccDNA formation.
