The present invention relates to a pharmaceutical composition and a health functional food comprising an extract of Korean rice wine lees as an active ingredient for prevention or treatment of thrombosis and, more specifically, to a pharmaceutical composition and a health functional food comprising, as an active ingredient, an ethanol extract or a hot water extract of Korean rice wine lees, or a mixture thereof, particularly, an ethyl acetate fraction, a butanol fraction, or a water residue of the extract of Korean rice wine lees, or a mixture thereof, for prevention or treatment of thrombosis. The extracts of Korean rice wine lees as an active ingredient of the pharmaceutical composition and the health functional food of the present invention are prepared by performing extraction on Korean rice wine lees using ethanol or the like to prepare an extract and then performing sequential fraction on the extract using hexene, ethyl acetate, and butanol. These extracts have an effect of efficiently inhibiting antithrombotic activity, antiplatelet activity, and thrombosis, due to the direct inhibition of human thrombin, and has an excellent effect of preventing or treating thrombosis, such as ischemic stroke and hemorrhagic stroke, by improving blood circulation. In particular, the extract of Korean rice wine lees of the present invention does not show acute oral toxicity, has excellent thermal stability, and does not show losses due to the direct inhibition of human thrombin, inhibition of prothrombin, and inhibition of blood clotting factors, even in acidic conditions of pH 2 and in the plasma.
