The current invention explores a novelistic approach dual drug delivery approach for colon disease. This approach was achieved by selecting an API and coupled with Nimesulide under controlled experimental conditions and the azo adduct was recrystallised by using methanol as solvent and subjected to invitro antibacterial activity by Disc Diffusion method. Muller Hinton was used as medium for bacterial strains. The azo adduct was evaluated for its various physicochemical parameters like color, solubility, Rf value, and melting point. The azo adduct formation was confirmed by IR and HNMR analysis. It was further subjected for evaluating its colon targeting property by in-vitro method using rat fecal matter. Tetracycline Nimesulide azo adduct tablets were formulated by using coprocessing agents like MCC,Starch, Magnesium stearate and talc and followed by direct compression. Significant drug release was achieved in colonic conditions for 24 Hrs and it also shows promising antibacterial activity upon the action of azo reductase enzyme. The same was performed by invitro agar by Disc Diffusion method. This novelistic approach is very effective and helps in minimizing the drug dosing frequency.
