The present invention relates to a method for producing a three-dimensional cultured skin model including a dermis and an epidermis, which comprises the following steps: preparing the dermis by using a composition including mouse-derived fibroblasts, and native collagen or a combination of the native collagen and atelocollagen； and forming the epidermis by using keratinocytes. In addition, the present invention relates to a three-dimensional cultured skin model including a dermis made of the composition comprising mouse-derived fibroblasts, and native collagen or a combination of the native collagen and the atelocollagen, and an epidermis formed by the keratinocytes. The three-dimensional cultured skin model of the present invention is excellent in formation and differentiation of a dermis and an epidermis, and has a structure similar to a natural skin layer by inhibiting collagen shrinkage and degradation phenomenon in the dermis by using a mouse-derived 3T3 cell and a mixture of the atelocollagen and the native collagen when producing the three-dimensional cultured skin model, thereby being widely used in fields of tests to evaluate toxicity and efficacy of drugs or cosmetics and alternative experiments of animal experiments.본 발명은 마우스 유래 섬유아세포； 천연형 콜라겐(Native collagen) 또는 천연형 콜라겐과 아텔로콜라겐(Atelocollagen)의 조합；을 포함하는 조성물을 이용하여 진피층을 제조하는 단계 및 케라틴 세포를 이용하여 표피층을 형성하는 단계를 포함하는 진피층 및 표피층을 포함하는 삼차원 배양 피부모델 제조방법에 관한 것이다. 또한, 본 발명은 마우스 유래 섬유아세포； 천연형 콜라겐 또는 천연형 콜라겐과 아텔로콜라겐의 조합；을 포함하는 조성물로 제조된 진피층과 케라틴 세포에 의해 형성된 표피층을 포함하는 삼차원 배양 피부모델에 관한 것이다. 본 발명의 삼차원 배양 피부모델은 제조시 마우스 유래 3T3 세포를 이용하고, 아텔로콜라겐과 천연형 콜라겐을 혼합하여 사용하는 것을 통하여 진피층과 표피층의 형성 및 분화가 우수하고 진피층의 콜라겐 수축 및 분해 현상을 억제하여 자연피부층에 유사한 구조를 가짐에 따라, 의약품이나 화장품의 독성 및 효능 실험, 동물실험 대체 실험 분야에서 널리 사용될 수 있다.
