A pharmaceutical composition for use in the treatment of headache comprising 4-9 parts by weight of Radix Angelicae Sinensis, 4-9 parts by weight of Rhizoma Chuanxiong, 2-8 parts by weight of Radix Paeoniae alba, 2-8 parts by weight of Radix Rehmanniae Preparata, 10-15 parts by weight of Ramulus Uncariae cum Uncis, 10-15 parts by weight of Caulis Spatholobi, 10-15 parts by weight of Spica Prunellae, 10-15 parts by weight of Semen Cassiae, 10 -15 parts by weight of Concha Margaritifera Usta, 4-9 parts by weight of Rhizoma Corydalis and 0.5-2 parts by weight of Herba Asari, characterized in that said composition is prepared by a method as follows: a). Preparation of extract No. 1: Radix Angelicae Sinensis, Rhizoma Chuanxiong, Rhizoma Corydalis and Semen Cassiae are mixed, extracted by refluxing with ethanol and filtered to remove impurities； and ethanol is recovered and concentrated, to give extract No. 1 for later use； b). Preparation of extract No. 2: Radix Paeoniae alba is extracted by refluxing with ethanol and filtered； and ethanol is recovered and concentrated, to give extract No. 2 for later use； C). Preparation of extract No. 3: Radix Rehmanniae Preparata, Ramulus Uncariae cum Uncis, Caulis Spatholobi, Spica Prunellae, Concha Margaritifera Usta and Herba Asari are mixed, subjected to decoction with water, filtered, concentrated, to which is added ethanol to let stand, and filter； and ethanol is recovered and concentrated to give extract # 3 for later use； d).
