Provided herein are compositions, methods, systems, and kits for wound healing. As shown herein, CCN2/CTGF stimulated mesenchymal progenitor cells can form &alpha；SMA&minus； fibroblasts. Further, TGF&bgr； was shown to stimulate further differentiation of &alpha；SMA&minus； fibroblasts to myofibroblasts associated with fibrosis. One aspect provides a composition including CCN2/CTGF and a TGF&bgr； inhibitor, a P38 inhibitor, or a tyrosine kinase inhibitor. Another aspect provides a method of treating tissue wounds with CCN2/CTGF-containing compositions. Also provided are systems and kits for wound healing. Also provided are methods for forming &alpha；SMA&minus； fibroblasts mesenchymal progenitor cells.
