The present invention relates to a medium composition for producing in vitro culture seedling and seminal roots of Rehmannia glutinosa and a mass production method of Rehmannia glutinosa using the same. More specifically, five types of mediums are in vitro cultured and a medium having an excellent growth rate is selected. Additionally, an auxin-based plant growth regulator is optimized by culture step and an auxin-based plant growth regulator, which forms multi-entity well and is capable of mass producing culture seedling of Rehmannia glutinosa, is selected. The in vitro culture seedling of Rehmannia glutinosa produced from the composition and the method of the present invention has bulky roots which are part used for medicinal purposes, optimizes soil acclimatization as an embryoid body of Rehmannia glutinosa, does not get sick easily and grows well.본 발명은 지황 기내 배양묘 및 종근 생산용 배지 조성물 및 이를 이용한 지황의 대량 생산방법에 관한 것으로, 상세하게는 5종의 배지를 기내 배양하여 생장율이 우수한 배지를 선정하였고, 추가로 옥신계 식물생장조절제를 배양 단계별로 최적화하여 다개체 형성이 잘되며 지황 배양묘를 대량 생산이 가능하게 하는 옥신계 식물생장조절제를 선정하였다. 본 발명에 따른 조성물 및 방법으로 생산된 지황 기내 배양묘는 약용부위인 뿌리가 비대할 뿐만 아니라, 지황 배양체로서 토양 순화가 잘되고, 쉽게 병에 걸리지 않으며, 생육이 잘 되는 효과가 있다.
