Provided herein is a fluid passage connection assembly for connecting fluid flow paths in a drug delivery device, eg, for delivery of a drug to a target site. The fluid passage connection assembly includes a hollow needle piercing member, a sleeve that is non-movably engaged with the piercing member, and a pierceable seal that is configured to sealingly engage the piercing member or sleeve. Upon sealing engagement, the piercing member or sleeve and the pierceable seal form a volume with a portion of the piercing member disposed therein. The fluid passage connection assembly is activated by relative movement between the piercing member and the pierceable seal, and activation causes the piercing member to pierce the pierceable seal, thereby establishing a fluid flow path through the pierceable seal. Upon assembly of the assembly into the sealed fluid flow path, the volume is separated from the environment and the sterility of the portion of the piercing member disposed therein is maintained.
