Disclosed herein is a method for ex vivo expanding tumor-infiltrating lymphocytes for use in adoptive cell therapy (ACT). The method involves culturing tumor fragments from the subject in a culture medium containing IL-2 and a 41BB agonist in an amount effective to expand tumor-infiltrating lymphocytes with enriched tumor-reactivity and specificity. Also disclosed is a method for treating a tumor in a subject that involves treating the subject with nonmyeloablative lymphodepleting chemotherapy, and administering tumor-infiltrating lymphocytes expanded by the disclosed methods.Linvention concerne un procédé dexpansion ex vivo des lymphocytes infiltrants des tumeurs, destinés à être utilisés dans le cadre de la thérapie cellulaire adoptive (ACT). Le procédé consiste à cultiver des fragments de tumeurs provenant du sujet, dans un milieu de culture contenant les agonistes IL-2 et 41 BB, dans une quantité efficace pour amplifier des lymphocytes infiltrant les tumeurs ayant une réactivité et une spécificité tumorales améliorées. Linvention concerne également un procédé de traitement dune tumeur chez un sujet qui consiste à traiter ce sujet par chimiothérapie non myéloablative appauvrissant les lymphocytes, et à administrer des lymphocytes infiltrant des tumeurs amplifiés par les procédés de linvention.
