The present invention relates to modified T cell receptors (TCRs) and to their use in adoptive cell therapy (ACT), in particular for the transfer of T lymphocytes. The TCRs are mutated in the transmembrane regions of the alpha and beta chains with mutations favoring the correct TCR chain pairing. The correct pairing of the transferred exogenous alpha and beta TCR chains improves the functional activity and safety of the genetically modified T cells for the therapy of tumours and infectious diseases. The invention also relates to T cell receptor alpha or beta chain, to a recombinant TCR, a TCR complex, a nucleic acid coding for the TCR alpha or beta chain, to relative recombinant expression vector, host cells, pharmaceutical composition and to a method of detecting a hematological malignant cell, a solid tumor cell or an infected cell.La présente invention concerne des récepteurs de lymphocytes T (TCR) modifiés et leur utilisation en thérapie cellulaire adoptive (ACT), en particulier pour le transfert de lymphocytes T. Les TCR sont mutés dans les régions transmembranaires des chaînes alpha et bêta avec des mutations favorisant lappariement correct des chaînes du TCR. Lappariement correct des chaînes alpha et bêta exogènes transférées du TCR améliore lactivité fonctionnelle et la sûreté en ce qui concerne les lymphocytes T génétiquement modifiés pour la thérapie des tumeurs et des maladies infectieuses. Linvention concerne également la chaîne alpha ou bêta du récepteur des cellules T, un TCR recombinant, un complexe TCR, un acide nucléique codant pour la chaîne alpha ou bêta du TCR, un vecteur dexpression recombinant relatif, des cellules hôtes, une composition pharmaceutique et un procédé de détection dune cellule hématologique maligne, une cellule de tumeur solide ou une cellule infectée.
