PROBLEM TO BE SOLVED: To provide methods and compositions for treating disorders affecting the central nervous system.SOLUTION: These disorders include neurometabolic disorders such as lysosomal storage diseases that affect the central nervous system, e.g., Niemann-Pick A disease. They also include disorders such as Alzheimer's disease. The methods for treating the disorders involve injecting to an axonal ending of a neuron a composition containing high titer adeno-associated virus (AAV) carrying a therapeutic transgene so that the AAV vector is transported in a retrograde fashion and transgene product is expressed distally to the administration site.【課題】中枢神経系に影響する疾患を治療するための方法および組成物の提供。【解決手段】中枢神経系に影響するリソソーム蓄積症のごとき神経代謝性疾患、例えば、ニーマン－ピック病Ａ型を含む。た、アルツハイマー病のごとき疾患を含む。疾患の治療法として、神経の軸索末端に治療用の導入遺伝子を有する高力価のアデノ随伴ウイルス（ＡＡＶ）を含む組成物を注入することにより、ＡＡＶベクターが逆行性様式で移送され、導入遺伝子の産物を投与部位より遠位で発現する方法。【選択図】なし
