MODULATOR OF MESENCHYMAL STEM CELL DIFFERENTIATION, MEDICAMENT USING THE SAME, AND METHOD OF SCREENING FOR SUBSTANCE HAVING MODULATING AND DIFFERENTIATING ACTIVITY TO MESENCHYMAL CELL
PROBLEM TO BE SOLVED: To provide a novel agent for modulating differentiation of mesenchymal cells, medicaments using the same, and a method of screening for pharmaceutical candidates.SOLUTION: The modulator of mesenchymal stem cell differentiation includes one or more selected from the group consisting of miRNA-294, a precursor thereof, and DNA constructs coding them as active ingredients, and promotes differentiation of mesenchymal cells. The medicaments including the differentiation modulator are also provided. The method of screening for substances having modulating and differentiating activity to mesenchyma cells includes: a process of supplying an analyte to mesenchymal stem cells, mesenchymal precursor cells, or a non-human animal expressing the miRNA-294; and a process of comparing the expression/functions of the miRNA-294 in the mesenchymal stem cells, the mesenchymal precursor cells, or the non-human animal to the expression/functions of the miRNA-294 during the period when the analyte is not supplied.【課題】間葉系細胞の分化を調節するための新規な剤、これを用いた医薬、並びに該医薬候補のスクリーニング方法の提供。【解決手段】miRNA-294、その前駆体およびこれらをコードするDNA構築物からなる群から選択される1種または2種以上を有効成分として含有し、間葉系細胞の分化を促進する、間葉系細胞の分化調節剤。上記分化調節剤を含む、医薬。miRNA-294を発現している間葉系幹細胞もしくは間葉系前駆細胞または非ヒト動物に対して被験物質を供給する工程と、前記間葉系幹細胞もしくは間葉系前駆細胞または非ヒト動物におけるmiRNA-294の発現または機能を、前記被験物質の非供給時におけるmiRNA-294の発現または機能と対比する、間葉系細胞への分化調節作用を有する物質のスクリーニング方法。【選択図】図2
