This disclosure provides ASCT2 binding molecules, such as anti-ASCT2 antibodies and antigen-binding fragments thereof. In some embodiments, such ASCT2 binding molecules are conjugated to a cytotoxic agent. For example, an ASCT2 antibody drug conjugate (ADC). In some embodiments, such anti-ASCT2 antibodies and fragments thereof may be hybridoma-derived murine monoclonal antibodies and humanized versions thereof. In some embodiments, such ASCT2 binding molecules specifically bind to cells that express ASCT2 and, in some cases, are internalized into cells. The present disclosure also provides compositions and methods for diagnosing and treating diseases or disorders characterized by ASCT2 overexpression, such as some types of cancer.
