The present invention relates to a siRNA transmitter comprising mesoporous silica nanoparticles (MSN) having fine pores with an average diameter of 5 nm or less and expanded pores of 10 to 50 nm, and siRNA coupled to outer surface of the particles and pore inner surface, wherein pore inner surface and outer surface of the particles comprise a first metal ion and a second ion, so that siRNA is coupled by using the same as a medium. In addition, the present invention relates to a method for manufacturing the siRNA transmitter and a pharmaceutical composition for treating cancer comprising the siRNA transmitter. The siRNA transmitter of the present invention exhibits excellent biocompatibility and can stably deliver siRNA into cells.본 발명은 평균 직경 5 nm 이하의 미세 기공 및 10 내지 50 nm의 확장된 기공을 갖는 메조기공 실리카 나노입자(mesoporous silica nanoparticle, MSN) 및 상기 입자의 외부 표면 및 기공 내부 표면에 결합된 siRNA를 포함하는 siRNA 전달체로서, 상기 입자의 기공 내부 표면 및 외부 표면은 제1금속이온과 제2금속이온을 포함하여 이를 매개로 siRNA가 결합되는 것인, siRNA 전달체에 관한 것이다. 또한, 본 발명은 상기 siRNA 전달체의 제조방법, 및 상기 siRNA 전달체를 포함하는 암 치료용 약학적 조성물에 관한 것이다.본 발명에 따른 siRNA 전달체는, 생물학적으로 안전한 칼슘 이온 등의 2가 금속 양이온을 사용하여, 종래의, 세포 독성을 가지므로 부작용을 유발할 수 있는, 아민을 이용한 기능화 없이 MSN 상에 siRNA를 로딩할 수 있다. 따라서, 본 발명의 siRNA 전달체는 우수한 생체적합성을 나타내며, siRNA를 안정적으로 세포 내로 전달할 수 있고, 공극 내에 추가의 약물을 담지함으로써, 유전자 및 약물을 동시에 전달하여 시너지적인 효과를 발휘할 수 있으므로, 암 치료를 위한 약학적 조성물로 이용될 수 있다.또한, 본 발명의 siRNA 전달체는 siRNA, 제1금속이온 함유 메조기공 실리카 나노입자 및 제2금속이온의 염을 단순 혼합하는 방법에 의해, 간편하고 원 포트로 제조될 수 있으며, 본 발명의 siRNA 전달체는 공극 및 입자의 크기, 및 표면 조성 등의 측면에서 다양한 성질을 갖는 넓은 범위의 실리카 입자에 대하여 확장될 수 있으므로, 신규한 유형의 siRNA 전달체 및 제조방법을 제공할 수 있다.
