PURPOSE:To obtain the title antibody useful for measuring reagent to detect carnations infected with MOTORU virus rapidly and simply, belonging to a class of IgM, produced by a hybridoma prepared from the virus as an immune source. CONSTITUTION:Dianthus barbatus L. raised into a seedling in a green house is transplanted to a planter. After two months, a carnation MOTORU virus (CarMV) is inoculated to the planter by carborundui method. After six weeks, leaves severe to a disease symptom are collected, a virus is separated by a conventional method, the virus and an adjuvant are administered to a Balb/c mouse and the mouse is immunized. After the final immunization, a splenic cell is collected, mixed with a mouse myeloma cell, fused by an electric fusion device and selectively cultured in a HAT medium to give a hybridoma. The hybridoma is cloned by limiting dilution and cultured to give the objective monoclonal antibody having a class (subclass) of IgM (K), showing positive reaction to an antigenic COUT protein which is derived from CarMV, reduced and has about 38,000 molecular weight.
