Benzocyclooctyne compounds of formula (I) are provided. These compounds undergo strain enhanced azide-alkyne cycloaddition (SPAAC) in the absence of toxic metal catalysts. The provided compounds are useful for diagnosis and imaging of azide-containing molecules. Disclosed are methods for detecting and imaging biomolecules using the compounds of the present disclosure.
