Disclosed herein are devices, instruments, systems, methods and kits for collecting and storing fluid samples from a subject. A device for collecting a fluid sample includes a housing including a recess having an opening, a vacuum chamber within the housing and in fluid communication with the recess, one that can extend through the opening to penetrate the skin of the subject. The puncture element described above can be included. The vacuum chamber can be configured to have a vacuum that pulls the skin into the recess. The recess can be configured to have a size or shape that allows an increased amount of fluid sample to accumulate in the skin that is drawn into the recess.
