The present invention relates to a composition for DNA targeting using a CRISPR interference (CRISPRi) system specific for TERT promoter mutation, a composition for preventing or treating cancer using the system, a diagnostic and imaging composition, a single strand guide RNA (sgRNA) used in the CRISPRi system, and a use thereof. According to the present invention, the single strand guide RNA (sgRNA) and the CRISPRi system using the same selectively and effectively target a point mutation of a TERT promoter and inhibit activities thereof. Targeting the -124 C>；T or -146 C>；T mutation TERT promoter through such epigenetic editing technique is expected to be highly suitable as a novel cancer therapy.본 발명은 TERT 프로모터 돌연변이에 특이적인 CRISPR interference (CRISPRi) 시스템을 이용한 DNA 표적용 조성물, 상기 시스템을 이용한 암의 예방 또는 치료용 조성물, 진단용 및 이미징 조성물, CRISPR interference (CRISPRi) 시스템에 이용되는 단일 가닥 가이드 RNA (sgRNA) 및 그의 이용에 관한 것이다. 본 발명에 따른 단일 가닥 가이드 RNA (sgRNA) 및 이를 이용한 CRISPRi 시스템은 선택적이고 효과적으로 TERT 프로모터의 점돌연변이 (point mutation)을 표적하고 이의 활성을 억제할 수 있는 바, 이러한 후성편집기술을 통한 -124 C>；T 또는 -146 C>；T 돌연변이 TERT 프로모터 표적화는 새로운 암 치료법으로써 매우 적합할 것으로 기대된다.
