Disclosed are: a safer live smallpox vaccine which contains an MSP strain in a reduced amount; and a method for production of the live smallpox vaccine. Specifically, disclosed is a method for production of a live smallpox vaccine, comprising the steps of: inoculating a master seed solution of attenuated vaccinia virus to a proper number of groups (1 to n; n is an integer) of rabbit primary kidney cells and culturing the cells; inoculating a part of the culture of each of the group to a RK-13 cell and a Vero cell, culturing the cells, and selecting and collecting a culture of a group which causes the formation of a plaque in a RK-13 cell but does not cause the formation of any plaque in a Vero cell; and preparing a vaccine stock solution using the culture (a working seed solution). Also specifically disclosed is a live smallpox vaccine prepared by the method.
