Provided is a method of treating or preventing age-related macular degeneration (AMD) in a patient subject to, or symptomatic of the disease, wherein the method comprises restoring normal lysosomal pH (pHL), or acidifying an abnormally elevated pHL, thus decreasing or preventing a damaging accumulation of lipofuscin or waste products in the retinal pigment epithelium (RPE) cells of the eye of the patient. Further, this method is achieved by modulating the action of the P2X7 and/or P2Y12 receptors of the RPE cells, specifically decreasing the acidity (pHL) of the RPE lysosomes by administering selected receptor antagonists affecting the action of the P2X7 and/or P2Y12 receptors of the RPE. Methods for selecting and quantifying the effectiveness of drugs to restore pHL and determine outer segment clearance rates is also provided using a high through-put screening protocol.Linvention concerne une méthode de traitement ou de prévention dune dégénérescence maculaire liée à lâge (AMD) chez un patient atteint ou symptomatique de la maladie, la méthode comprenant la restauration dun pH lysosomal normal (pHL) ou lacidification dun pHL anormalement élevé, diminuant ainsi ou empêchant une accumulation détérioratrice de lipofuscine ou de produits de déchets dans les cellules de lépithélium pigmentaire rétinien (RPE) de lœil du patient. En outre, cette méthode est obtenue par la modulation de laction des récepteurs P2X7 et/ou P2Y12 des cellules RPE, en particulier la diminution de lacidité (pHL) des lysosomes du RPE par ladministration dantagonistes choisis des récepteurs affectant laction des récepteurs P2X7 et/ou P2Y12 du RPE. Linvention concerne également des procédés de sélection et de quantification de lefficacité de médicaments pour restaurer le pHL et déterminer les vitesses de clairance du segment externe à laide dun protocole de criblage à haut débit.
