Substrates, optionally coated with an undercoating layer, having grafted therefrom one or more non-fouling materialsare described herein. The non-fouling, polymeric material can be grafted froma variety of substrate materials, particularlypolymeric substrates and/or polymeric undercoating layers. The graft-fromtechniques described herein can result in higher surfacedensities of the non-fouling material relative to graft-to formulations. Graft-from methods can be used to produce covalently tetheredpolymers. The compositions described herein are highly resistant proteinabsorption, particularly in complex media and retaina high degree of non-fouling activity over long periods of time. Thecompositions described herein may also demonstrate antimicrobialand/or anti-thrombogenic activity. The non-fouling material can be graftedfrom the substrate, or optionally from an undercoatinglayer on the substrate, preferably without significantly affecting themechanical and/or physical properties of the substratematerial.
