The present invention relates to a method for culturing tetraploid balloon flower roots using in vitro culture and a method for preparing a medium for inducing adventitious roots using the same. The method for culturing tetraploid balloon flower roots using in vitro culture and the method for preparing a medium for inducing adventitious roots using the same comprise: (a) a step for immersing balloon flower roots cultured in vitro in a colchicine solution and suspension culturing in the darkness (b) a step for washing the suspension cultured balloon flower roots with sterilized water and seeding on a 1/2 MS medium and (c) a step for culturing the germinated plant bodies of the balloon flower roots in the 1/2 MS medium again. The method for culturing tetraploid balloon flower roots using in vitro culture and the method for preparing a medium for inducing adventitious roots using the same of the present invention enable the mass production of balloon flower roots more effectively compared to the conventional methods, thereby being able to contribute to an increase in rural household incomes.COPYRIGHT KIPO 2015본 발명은 기내배양을 이용한 4배체 도라지의 배양방법 및 이를 이용한 부정근 유도를 위한 배지 제조 방법에 관한 것이다. 본 발명의 기내배양을 이용한 4배체 도라지의 배양방법 및 이를 이용한 부정근 유도를 위한 배지 제조 방법은 (a)기내 배양한 도라지를 콜히친 용액에 침지하여 암 상태에서 현탁 배양하는 단계 (b) 현탁 배양한 도라지는 멸균수로 세척하고 1/2 MS 배지에 치상하는 단계 및 (c) 발아된 도라지 식물체를 1/2 MS 배지에 다시 배양하는 단계를 포함하여 이루어지는 것을 특징으로 한다. 본 발명의 기내배양을 이용한 4배체 도라지의 배양방법 및 이를 이용한 부정근 유도를 위한 배지 제조 방법은 종래 방법에 비해 효과적으로 도라지를 대량생산할 수 있어 농가의 소득 향상에 기여할 수 있는 효과가 있다.
