A transfer device (2) for handling fluids in a sterile manner, comprising: a body (4) having an bore (10) formed through at least a part of its interior, the body (4) has a first end (6) connectable to an upstream component (54; 72); an opening (62) located downstream of the first end (6), the opening can be connected to a downstream component (56; 64; 210); a plunger (18; 70; 98; 108) contained within the bore, the plunger has a first end and is movable within the bore (10), by movement of a cam (32) in a cam groove (36) which defines an axial movement and a rotation and that limits the length of movement of the piston in the bore, between a closed position corresponding to a closed position of the device in which the first end (6) of the body (4) is hermetically closed with respect to the opening (62) by means of a first hermetic seal (38) and an open position corresponding to an open position of the device in which the first end (6) of the body and the opening are connected with fluid transfer, where, when the plunger diver (18; 70; 98; 108) is in the closed position, the first end of the plunger (18; 70; 98; 108) combined with the first end (6) of the body (4) form a surface (46 ; 104) configured to be sterilized with water vapor and a barrier is between the environment and at least a part of the interior of the body (4) and the plunger (18; 70; 98; 108); a fluid path (27; 74) forming a fluid connection through the device between the first end (6) and the opening (62) when the device is in the open position; and an additional seal comprising two seals (38) or a linear or gland seal (90) arranged along the plunger (18; 70; 98; 108) to form a liquid-tight seal between various parts of the plunger (18; 70; 98; 108) and bore (10) when the plunger (18; 70; 98; 108) is in the open position to allow fluid transfer through the fluid path while maintaining the sterile state of the fluid being transferred as long as a sterile connection is maintained with the upstream com
