The present invention relates to a method for supercritical extraction of 2-hydroxy-4-isopropyl-2,4,6-cycloheptatrien-1-one derived from Japanese cypress and to a composition containing the same for treating atopic dermatitis. The method for extraction of the Japanese cypress-derived 2-hydroxy-4-isopropyl-2,4,6-cycloheptatrien-1-one comprises (a) a step for drying the wood of Japanese cypress using cool air (b) a step for smashing the dried wood of Japanese cypress to 0.2-2 mm of splinters (c) a step for performing a first supercritical extraction of the splinters of the wood of Japanese cypress using carbon dioxide under temperatures of 30-50°C and pressures of 100-500 bar (d) a step for performing a second supercritical extraction of a first supercritical extract of the wood of Japanese cypress using carbon dioxide under temperatures of 30-50°C and pressures of 100-500 bar and (e) a step for extracting 2-hydroxy-4-isopropyl-2,4,6-cycloheptatrien-1-one from the a second supercritical extract of the wood of Japanese cypress.COPYRIGHT KIPO 2015본 발명은 편백 유래 2-하이드록시-4-이소프로필-2,4,6-싸이클로헵타트리엔-1-원(2-hydroxy-4-isopropyl-2,4,6-cycloheptatrien-1-one)의 초임계 분리 방법 및 이를 포함하는 아토피성 피부염 치료용 조성물에 관한 것이다.상기 편백 유래 2-하이드록시-4-이소프로필-2,4,6-싸이클로헵타트리엔-1-원의 분리 방법은 (a) 편백나무 목질부를 냉풍 건조하는 단계 (b) 상기 건조된 편백나무 목질부를 0.2 내지 2 ㎜의 입자 크기로 분쇄하는 단계 (c) 상기 편백나무 목질부 분쇄물을 30~50℃의 온도 및 100~500bar의 압력 하에서 초임계 이산화탄소로 1차 초임계 추출하는 단계 (d) 상기 편백나무 목질부의 1차 초임계 추출물을 30~50℃의 온도 및 100~500bar의 압력 하에서 초임계 이산화탄소로 2차 초임계 추출하는 단계 및 (e) 상기 편백나무 목질부의 2차 초임계 추출물로부터 2-하이드록시-4-이소프로필-2,4,6-싸이클로헵타트리엔-1-원을 분리하는 단계를 포함한다.
