The successful development of a novel resveratrol and doxorubicin loaded liposomal nanoparticle and evaluation of its in vitro and in vivo activity is described in this study. The combination of resveratrol and doxorubicin for the treatment of head and neck cancer has been explored for the first time. The characterization of the nanoparticle shows that the drug localization and their interaction with the liposomal membrane influence other parameters like drug encapsulation and release. The in vitro efficacy was evaluated on two head and neck squamous cell carcinoma (HNSCC) cell lines NT8e and AW8507 it was observed that the drug combination encapsulated in the liposomal formulation showed better efficacy than the free drugs. The evaluation of molecular pathways showed that the drug- loaded nanoparticle exerts apoptosis inducing effects by controlling the cell cycle and downstream apoptosis proteins. In vivo evaluation of this formulation showed cell cycle arrest at G2/M transition after a treatment of a weekly dose for 1 month. Also, nanoencapsulation of the drug combination successfully reduced locoregional skin toxicity and helped in maintaining gross body weight of the treated animals when compared to the severe weight loss and morbidity experienced by the free drug treated animals. This work suggests that development of dual drug delivery nanosystems for drug combinations holds much promise in the treatment of HNSCC.
