The invention provides a method for obtaining a broadly neutralizing antibody(bNab), including screeningmemory B cell cultures from a donor PBMC sample for neutralization activityagainst a plurality of HIV-I species, cloning a memory Bcell that exhibits broad neutralization activity; and rescuing a monoclonalantibody from that memory B cell culture. The resultantmonoclonal antibodies are characterized by their ability to selectively bindepitopes from the Env proteins in native or monomericform, as well as to inhibit infection of HIV-I species from a plurality ofclades. Compositions containing human monoclonalanti-HIV antibodies used for prophylaxis, diagnosis and treatment of HIVinfection are provided. Methods for generating suchantibodies by immunization using epitopes from conserved regions within thevariable loops of gp120 are provided. Immunogens forgenerating anti-HIV 1 bNAbs are also provided. Furthermore, methods forvaccination using suitable epitopes are provided.
