Anti-SIRPα antibodies, including multispecific anti-SIRPα antibodies, are provided as well as related compositions and methods. The antibodies of the present disclosure bind to SIRPα and block the interaction between CD47 on one cell and SIRPα on phagocytic cells. The subject anti-SIRPα antibodies find use in a variety of therapies. Embodiments of the present disclosure include isolated antibodies and derivatives and fragments thereof, pharmaceutical formulations comprising one or more anti-SIRPα antibodies, and cell lines that produce the antibodies.
