PROBLEM TO BE SOLVED: To provide a modified biotin-binding protein which can be expressed in a soluble form in a high yield in bacteria. Also provided is a fusion protein comprising a modified biotin-binding protein and an antigen. A soluble biotin-binding protein comprising a specific amino acid sequence and/or any functional derivative thereof. Also, a non-hemolytic variant of α-hemolysin from Staphylococcus aureus, and a fusion protein comprising a non-hemolytic variant of α-hemolysin and a biotin-binding domain. The immunogenic composition containing the protein is used to induce an immune response or to vaccinate a subject.
