The present invention relates to a large quantity production of Rg3 (r form) which is one among water-insoluble saponin, from the leaves and roots of ginseng naturally grown domestically. More particularly, the large quantity production of Rg3 comprises: a ginseng material pre-treatment step of washing the roots or leaves of ginseng, naturally drying the same, and pulverizing the resultant product a ginseng saponin extraction and conversion step of converting water-soluble saponin Rg3(s) into water-insoluble saponin Rg3(r) by obtaining an extract by adding alcohol to the pulverized material, removing chlorophyll, and passing the resultant product through a high temperature and high pressure treatment, frying treatment, and enzyme treatment and a step of recovering and purifying water-insoluble saponin Rg3(r) having high purity and white crystals by obtaining a ginseng extract precipitated by diluting a ginseng extract with water, obtaining a water-insoluble saponin fraction by using the resultant product and MPLC, splitting water-insoluble saponin Rg3(r) by using HPLC, and by freeze-drying the resultant product. The present invention relates to a technique to produce a large quantity of water-insoluble Rg3(r) as a material which can replace water-soluble Rg3(s) derived from ginseng, which is recently used as a raw material for an anti-cancer medicine.
