The present invention relates to a technique of regulating a whole body energy metabolism including liver by liver-specific regulation of Crtc2. A pharmaceutical composition for preventing, alleviating or treating obesity or fatty liver comprises, as an active ingredient, material which liver-specifically inhibits expression of a gene coding a Crtc2 (CREB regulated transcription coactivator 2) protein, or liver-specifically inhibits activity of the Crtc2 protein. More specifically, the pharmaceutical composition according to the present invention has effects of inhibiting blood-sugar elevation due to high fat diet and improving glucose and insulin resistance values by liver-specific inhibition of Crtc2. Further, the pharmaceutical composition according to the present invention reduces sizes and fat accumulation of fat cells in addition to liver cells by liver-specific inhibition of Crtc2, and has an anti-obesity effect through an increase in energy consumption. Further, the pharmaceutical composition according to the present invention has an effect of enabling the fatty liver to be suppressed since oxidation of fat is increased due to the liver-specific inhibition of Crtc2. Therefore, the liver-specific inhibition of Crtc2 of the present invention not only can be applied to prevention, alleviation or treatment of obesity, fatty liver and the like, but also can be used in developing a therapeutic agent of obesity, fatty liver and the like by using liver-specific regulation of Crtc2.본 발명은 Crtc2 간 특이적 조절에 의한 간 포함 전신체적 에너지 대사를 조절하는 기술에 관한 것이다. 보다 구체적으로, Crtc2 간 특이적 억제에 의해 고지방식이에 의한 혈당 상승이 억제되고, 포도당과 인슐린 저항성이 개선되는 효과가 있다. 또한, Crtc2 간 특이적 억제에 의해 간세포 이외에 지방세포의 크기 및 지방축적이 감소하며, 에너지 소모 증가를 통해, 항 비만 효과가 있다. 또한, Crtc2 간 특이적 억제에 의해 지방 산화가 증가되어 지방간을 억제할 수 있는 효과가 있다. 따라서, 본 발명의 Crtc2 간 특이적 억제는 비만 또는 지방간 등의 예방, 개선 또는 치료에 적용될 수 있으며, Crtc2 간 특이적 조절을 이용하여 비만 또는 지방간 등의 치료제 개발에도 이용될 수 있다.
