The present invention relates to a method of testing whether a patient suffering of multiple myeloma will respond or not to a DNA methyltransferase inhibitor (DNMTi) comprising: i. determining the expression level (ELi) of several genes GI-Gn selected from table A in a biological sample obtained from said patient ii. comparing the expression level (ELi) determined at step i) with a predetermined reference level (ELRi) iii. calculating the DMS score trough the following formula (I) wherein βί represent the regression β coefficient reference value for the gene Gi and Ci = 1 if the expression of the gene Gi (ELi) is higher than the predetermined reference level (ELRi) or Ci = -1 if the expression of the gene (ELi) is lower than or equal to the predetermined reference level (ELRi) comparing the score DMS determined at step iii) with a predetermined reference value DMSR and concluding that the patient will respond to the DNMTi when the DMS score is higher than the predetermined reference value DMSR or concluding that the patient will not respond to the DNMTi when the DMS score is lower than the predetermined reference value DMSR.La présente invention concerne un procédé permettant de tester si un patient souffrant de myélome multiple répondra ou non à un inhibiteur de la méthyltransférase de lADN (DNMTi) consistant à : i. déterminer le niveau dexpression (ELi) de plusieurs gènes GI-Gn choisis dans le tableau A dans un échantillon biologique obtenu dudit patient ii. comparer le niveau dexpression (ELi) déterminé à létape i) avec une niveau de référence prédéterminé (ELRi) iii. calculer le score DMS par le biais de la formule suivante (I) où βί représente la valeur de référence du coefficient de régression β du gène Gi et Ci = 1 si lexpression du gène Gi (ELi) est plus élevée que le niveau de référence prédéterminé (ELRi) ou Ci = -1 si lexpression du gène (ELi) est plus basse que le niveau de référence prédéterminé (ELRi) ou égale à celui-ci par rapport au score DMS déterm
