PURPOSE: An isolation and purification method of platicodi radix fractions including high purity platycoside is provided to simply and economically obtain platicodi radix fractions in short time. CONSTITUTION: An isolation and purification method of platicodi radix fractions including high purity platycoside comprises the following steps: dissolving platicoside ethanol crude extract in ethanol and adding acetone； removing precipitates and concentrating filtrate in order to obtain platicoside ethanol-acetone fraction； obtaining water fraction by dissolving the platicoside ethanol-acetone fraction and concentrating water layer by dividing ethylacetate； obtaining butanol fractions by concentrating the butanol layer after separting the water layer from the butanol layer； and isolation purifying the Platicodi Radix fractions which contains high purity Platicodi Radix platicoside by using reverse phase cartridge chromatography.본 발명은 고순도의 길경 프라티코사이드를 포함하는 길경 분획물의 분리정제 방법에 관한 것으로서, 더욱 상세하게는 1) 길경 에탄올 조추출물을 에탄올에 용해시키고 아세톤을 첨가하여 형성되는 침전물을 제거하고 여액을 농축하여 길경 에탄올-아세톤 분획을 수득하는 단계； 2) 상기 길경 에탄올-아세톤 분획을 물에 용해시키고 에틸아세테이트로 분배하여 물층을 농축하여 물 분획을 수득하는 단계； 3) 상기 2) 단계의 물층에 수포화 부탄올을 혼합하고 물층과 부탄올층으로 층분리한 후 부탄올층을 농축하여 부탄올 분획을 수득하는 단계； 및, 4) 상기 부탄올 분획을 증류수에 용해시키고 에탄올 함유 수용액을 용출용매로 하는 역상(C18) 카트릿지 크로마토그래피에 의해 길경 프라티코사이드를 고순도로 함유하는 길경 분획물을 분리 정제하는 단계를 포함하여 이루어지는 길경 에탄올 추출물로부터 프라티코사이드를 고순도로 함유하는 길경 분획물을 분리 정제하는 방법에 관한 것이다.본 발명에 의하면, 길경 사포닌인 프라티코딘 D(platycodin D)와 길경 프라티코사이드(platycosides)를 고순도 및 고농도로 함유하는 길경 분획물을 간단하고 경제적인 방법으로 단시간에 수득할 수 있는 효과를 기대할 수 있다.
