PROBLEM TO BE SOLVED: To solve a problem that, though a lipocalin family has a pocket with a barrel structure, in other words, a cup-like shape, and can store a hardly water-soluble medicine in the same, in addition, a plurality of molecule units can be stored depending on the medicine, the pocket formed in the barrel structure has always opening, therefore, there is a risk of dropping the medicine already having been included during conveyance.SOLUTION: In using a lipocalin family as a capsule for a DDS, a pocket opening is brought to be capable of being closed with a disulfide crosslinking (S-S bond) after storing a medicine in the pocket formed in a barrel structure. To be concrete, enzyme activity of a lipocalin type prostaglandin D synthetase is deactivated and each of tryptophan residues at 34th and 92th positions from an N-terminal is substituted with a cysteine residue.【課題】リポカリンファミリーはバレル構造によってポケットを有しており、いわばコップのような形状をしている。ここに、難水溶性の薬剤を収納することができる。しかも、薬剤によっては、複数個の分子単位を収納することができる。しかしながら、バレル構造によって形成されたポケットは、常に開口しており、移送の途中で内在させた薬物がこぼれてしまうおそれがある。【解決手段】リポカリンファミリーをＤＤＳ用のカプセルとして利用する際に、バレル構造で形成されたポケット中に薬物を収納した後、ポケットの開口をジスルフィド架橋（Ｓ－Ｓ結合）によって閉じることができるようにするものである。具体的には、リポカリン型プロスタグランジンＤ合成酵素の酵素活性を失活させ、Ｎ末端から３４番目と９２番目のトリプトファンをシステインに置き換えたことを特徴とするＤＤＳカプセル用タンパク質を提供する。【選択図】図２
