Disclosed are a biodegradable amphiphilic polymer with a side chain containing double sulfur, a self-crosslinking polymer vesicle thereof and a use in the targeted therapy of lung cancer. The polymer is obtained based on a cyclic carbonate monomer containing a double sulfur five-membered ring functional group through the actively controlled opening-ring and polymerization, wherein the molecular weight is controllable and the distribution of the molecular weight is relatively narrow, without the need for the processes of protection and deprotection. The polymer obtained through the opening-ring and polymerization of the cyclic carbonate monomer in the present invention has biodegradability, and can be used to control the drug release system. Reduction-sensitive and reversibly cross-linked vesicle nano-drug carriers targeted at lung cancer are prepared to support stable and long circulation in vivo, with high enrichment in the lung cancer tissues and efficiently entry into the cells, and can rapidly degrade the cross-link and release the medicine into the cells, efficiently and specifically kill the cancer cells, and effectively inhibit the growth of tumors without causing toxic side effects.
