The present invention relates to a method of mass propagating bamboo by using in vitro culture, and a medium composition for in vitro culture of bamboo. More specifically, the present invention relates to a method of mass propagating bamboo by using shoots of a temperate bamboo growing in a temperate zone； and a bamboo plant body produced thereby. The mass propagation method comprises: (a) extracting a node piece including an auxiliary bud from a bamboo branch and sterilizing the same； (b) culturing the sterilized node piece to generate a shoot； (c) sub-culturing the generated shoot to induce multiple shoots； (d) culturing the induced multiple shoots to induce rooting； (e) culturing the rooting-induced multiple shoots to grow roots and stems to thereby obtain a plantlet； and (f) subjecting the plantlet to sensory adaptation to grow the same into a normal bamboo plant body. Accordingly, the method of the present invention can mass propagate identical bamboo entities over a short period of time through in vitro culture, and by inducing rooting, can obtain normal plant bodies whenever the need arises, and thus can be extremely beneficially used in the mass propagation of bamboo.본 발명은 기내배양에 의한 대나무 대량증식 방법 및 대나무 기내배양용 배지 조성물에 관한 것이다. 더욱 상세하게는 온대지역에서 자라는 온대종 대나무의 신초를 이용한 대량증식 방법에 있어서, (a) 대나무의 가지에서 측아를 포함하는 마디 절편을 채취하여 멸균시키는 단계； (b) 상기 멸균한 마디 절편을 배양하여 신초를 발생시키는 단계； (c) 상기 발생한 신초를 계대배양하여 다발성 신초를 유도하는 단계； (d) 상기 유도된 다발성 신초를 배양하여 발근을 유도하는 단계； 및 (e) 상기 발근이 유도된 다발성 신초를 배양하여 뿌리와 줄기를 성장시켜 유식물체를 얻는 단계； (f) 상기 유식물체를 순화시킨 후 정상적인 대나무 식물체로 육성하는 단계를 포함하는 대나무 대량증식 방법 및 이에 의해 생산된 대나무 식물체에 관한 것이다. 상기 방법을 이용하면 기내배양을 통하여 짧은 기간에 동일한 대나무 개체를 대량으로 증식시키고 필요할 때마다 발근을 유도하여 정상적인 식물체를 얻을 수 있는 바, 대나무 대량증식에 매우 유용하게 사용될 수 있다.
