The present invention provides a varenicline formulation for controlled-release and a manufacturing method thereof, wherein the varenicline formulation comprises: a core containing varenicline or a pharmaceutically acceptable salt thereof as an active component； and a controlled release layer containing a plurality of active component eluting materials having different dissolution levels formed on the core. The controlled release layer comprises a first coating layer containing a first active component eluting material and a second coating layer comprising a second active component eluting material, wherein the first coating layer is formed between the core and the second coating layer, and a content ratio of the core, the first coating layer, and the second coating layer is 60 to 80 : 1 to 10 : 10 to 40.
