The present invention relates to pharmaceutical compositions useful for topical, non-invasive delivery of an oligonucleotide to the ear and to methods for the treatment of an ear disorder, including hearing loss arising from chemical-induced ototoxicity, acoustic trauma and presbycusis and microbial infections. The method comprises topically administering to the ear of a subject in need thereof a pharmaceutical composition comprising an inhibitory oligonucleotide, a permeability enhancer and a pharmaceutically acceptable carrier, wherein the oligonucleotide reduces or inhibits expression of a gene associated with the ear disorder in the subject.La présente invention concerne, dune part des compositions pharmaceutiques convenant à ladministration topique et non invasive dun oligonucléotide dans loreille, et dautre part des procédés pour le traitement dun trouble de loreille, notamment la perte daudition imputable à une ototoxicité chimio-induite, les traumatismes acoustiques et la presbyacousie, ainsi que des infections microbiennes. Le procédé consiste à administrer, dans loreille dun patient justifiant dun tel traitement, une composition pharmaceutique comprenant un oligonucléotide inhibiteur, un renforçateur de perméabilité, et un vecteur pharmaceutiquement admis, loligonucléotide réduisant ou bloquant lexpression dun gène associé au trouble affectant loreille du patient.
