The present invention relates to a bethanechol sustained-release formulation and a method for manufacturing the same. More specifically, the present invention relates to: the bethanechol sustained-release formulation for once a day oral administration, which contains (i) a matrix formulation formed from granules comprising betacol or a pharmaceutically acceptable salt, and a mixture of a swellable polymer and a water-insoluble polymer as active components, and (ii) a coating layer for coating the matrix formulation which contains the mixture of an insoluble polymer and a hydrophilic polymer； and the method for manufacturing the same. A sustained-release formulation for once a day oral administration according to the present invention forms a hydrogel while using a minimum amount of a polymer, so that bethanechol can be continuously controlled and released for 24 hours and it is possible to improve the convenience of taking by having a suitable size for taking.본 발명은 베타네콜 서방성 제제 및 이의 제조방법에 관한 것으로, 보다 상세하게는 (i) 활성성분으로서 베타네콜 또는 이의 약제학적으로 허용가능한 염, 및 팽윤성 고분자와 비수용성 고분자의 혼합물을 포함하는 과립물로부터 형성된 매트릭스 제제； 및 (ii) 상기 매트릭스 제제를 코팅하는 불용성 고분자와 친수성 고분자의 혼합물을 포함하는 코팅층을 포함하는 1일 1회 경구투여용 베타네콜 서방성 제제； 및 이의 제조방법에 관한 것이다. 본 발명에 따른 1일 1회 경구투여용 서방성 제제는 최소량의 고분자를 사용하면서도 수화겔을 형성하여, 24시간 지속적으로 베타네콜을 제어방출시킬 수 있고, 복용하기 적당한 크기를 가져 복용의 편리성을 개선할 수 있다.
