Useful compositions and methods of providing in vivo release of substantially water-insoluble pharmacologically active agents (such as the anticancer drug paclitaxel) wherein the pharmacologically active agent is released as suspended protein-coated particles are provided in accordance with the present invention. The procedure produces particles with a diameter of less than about 1 micron. The use of the specific composition and preparation conditions (eg addition of a polar solvent to the organic phase), and careful selection of the appropriate organic phase and phase fraction, enables the serial production of unusually small nanoparticles of less than 200 µm. nm in diameter, which can be sterile filtered. The particulate system produced according to the invention may be converted into a redispersible dry powder comprising a protein-coated, water-insoluble drug nanoparticles and a free protein to which the pharmacological agent molecules are adhered.
