The present invention relates to a method of treating cancer by in vivo priming and activation of natural killer cells to achieve tumor cell lysis. The method includes introducing into a patient a priming tumor cell preparation (PTCP) derived from a first tumor cell line, wherein the preparation is irradiated to inactivate the first tumor cell or membrane preparation thereof and the first Tumor cells have a known priming ligand on their membrane surface. The resting NK cells of the patient are contacted with PTCP in vivo, resulting in primed NK cells, which are characterized by upregulation of CD69, removal of CD16, or a combination of CD69 + and CD16−.
