Methods of efficiently producing antibodies and other multimeric protein complexes (collectively referred to herein as heteromultimer proteins) capable of specifically binding to more than one target are described herein. The target can be, for example, a different epitope on a single molecule, or it can be located on a different molecule. The method combines efficient high gene expression levels, suitability of assembly and ease of purification for heteromultimeric proteins.
