Multispecific binding proteins that bind to PSMA, NKG2D receptor, and CD16 are described, as well as pharmaceutical compositions and therapeutic methods useful for treating cancer. The multispecific binding protein can bind to PSMA in cancer cells or cancer neovasculature and NKG2D and CD16 receptors in natural killer cells and associate with two or more NK activating receptors. And the binding of the natural ligand to NKG2D.ＰＳＭＡ、ＮＫＧ２Ｄ受容体、およびＣＤ１６に結合する多重特異性結合タンパク質、ならびにがんの処置に有用な医薬組成物および治療方法が記載される。上記多重特異性結合タンパク質は、がん細胞またはがんの新生血管におけるＰＳＭＡならびにナチュラルキラー細胞におけるＮＫＧ２Ｄ受容体およびＣＤ１６受容体に結合し、２種類以上のＮＫ活性化受容体と会合することができ、そして天然リガンドのＮＫＧ２Ｄへの結合を阻止し得る。
