Glucagon-like peptide-1 (GLP-1) analogs for promoting secretion of insulin, named as truncated glucagon-like peptide-1 (sGLP-1), is composed of 26 amino acid residues with the sequence of His-X1-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Val-Ser-Ser-Tyr-Leu-Glu-Gly-Gln-Ala-Ala-Lys-Glu-Phe-Ile-Ala-Trp-Leu. The sGLP-1 according to the present invention has marked merits compared with current GLP-1 analogs, as follows: a) stronger binding affinity to insulin cell receptor and enhanced capacity to stimulate insulin secretion b) resistance to degradation of dipeptidyl peptidase because of the substitution of Gly or Ser for Ala at amino-acid position 2, resulting in longer in vivo half life and improved curative effect c) lowering synthetic cost due to shorter sequence.La présente invention a trait à des analogues de glucagon-like peptide-1 pour la promotion de la sécrétion dinsuline, désignés glucagon-like peptide-1 tronqué (sGLP-1) constitué de 26 résidus dacides aminés avec la séquence His-X1-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Val-Ser-Ser-Tyr-Leu-Gly-Gln-Ala-Lys-Glu-Phe-Ile-Ala-Trp-Leu. Le sGLP selon linvention présente des qualités remarquables par rapport aux analogues GLP-1 connus, notamment: a) un meilleure affinité de liaison au récepteur cellulaire de linsuline et une capacité améliorée pour la stimulation de la sécrétion de linsuline b) un résistance à la dégradation de dipeptyl peptidase grâce à la substitution de Gly ou Ser par Ala à la position 2 dacides aminés, entraînant une plus longue demi-vie in vivo et un effet curatif amélioré c) la réduction de coût de synthèse grâce à une séquence plus courte.
